Ever since their discovery in 1996, chemokine receptorswhich act as HIV coreceptors have been aninteresting new target for drug development. Thefirst class of CXCR4 targeted drugs, the non-peptidicbicyclams, were discovered in 1992 (De Clercqet al. PNAS 89:5286-90), long before its precisemechanism of action was unraveled (Schols et al. JExp Med 1997;186:1383-8).
