Schedules of drug administration are normallybased on the elimination half-life (t1/2) of the drugs.Medications with long t1/2 (> 24 hours), such as nonnucleosidereverse transcriptase inhibitors (NNRTI),can be dosed once a day. By contrast, most proteaseinhibitors (PI), nucleoside reverse transcriptaseinhibitors (NRTI), and integrase strand transfer inhibitors(InSTI) have shorter t1/2 and, therefore, aregenerally given twice a day. However, the particularpharmacodynamic and pharmacokinetic characteristicsof NRTI and InSTI may also support once-adaydosing.
